Protective Effect of Nervonic Acid Against 6-Hydroxydopamine-Induced Oxidative Stress in PC-12 Cells

Increased oxidative stress in the human brain is observed neurodegenerative diseases such as Parkinson’s disease (PD) and Alzheimer’s (AD), considered to be a major cause of progression these states. A very long-chain fatty acid, nervonic acid (NA), main found various sphingolipid species central nervous system. NA plays an important role forming plasma membrane’s lipid bilayer maintaining normal myelin function. In this study, we examined neuroprotective effect against rat pheochromocytoma (PC-12) cells stimulated by 6-hydroxydopamine (6-OHDA), which served cell model PD. PC-12 were pre-treated with different concentrations for 48 h then subsequently co-treated 6-OHDA induce cellular stress. Cell viability was significantly increased pre-treatment low concentration NA. The level malondialdehyde, marker peroxidation, decreased NA-treated cells. expression levels superoxide dismutases (Mn SOD Cu/Zn SOD) ?-glutamylcysteine synthetase (GCLC), responsible synthesis glutathione, increased, indicating that activated antioxidant defense These results suggest may play mediator brain.
